Effect of A20 on glucose dependent cell migration in acute lymphoblastic leukemia

Acute lymphoblastic leukemia (ALL) is the most common pediatric hematologic malignancy characterized by aberrant proliferation of immature lymphoid cells. A20 a deubiquitinase gene that inhibits functional activation immune cells mediated through nuclear factor κB (NFκB)/signal transducers and activators transcription (STAT) pathways. frequently inactivated in leukemia/lymphoma. Little known about involvement between STAT signalling regulating function ALL blasts. The present study, therefore, explored whether migration apoptosis peripheral blood mononuclear (PBMCs) blasts high glucose conditions regulated A20. To this end, from samples fifteen patients PBMCs healthy individuals absence were examined. Gene expression profile was determined quantitative RT-PCR, cell flow cytometry, transwell assay. As result, Cell migration, but not ALL-blasts enhanced when exposed to dependent on expression, effects abolished using Nifuroxazide, inhibitor. In conclusion, inhibited glucose-induced pathway. effect might contribute poorer survival patients, who develop hyperglycemia during therapy.
